Dengue virus RNA (RT-PCR)

What it measures
Detects dengue virus RNA and can identify the serotype (DENV-1 to 4). It is the most specific test in the first 5 days of fever.
Specimen and preparation
- Specimen: Serum or EDTA plasma
- Tube: Gold top
- Preparation: Collect within 5 days of fever onset; keep cold and send promptly.

Reference ranges differ between laboratories, methods and populations. Always use the range printed on your own lab report.
Causes of high values
- Acute dengue viremia
- Identifies the circulating serotype for surveillance
- Positive before NS1 or antibodies in some patients

Real-time RT-PCR (with serotyping)
Good to know
Viremia falls quickly after day 5, so a negative PCR later should be followed by IgM testing.
